It is unknown when she infiltrated HYDRA. Like the real Madame Hydra, she served as one of Baron Strucker's lieutenants. After the Breakout, Black Widow brought a sample of the Hulk's blood to her. She was about to pay her when Hawkeye attacked the HYDRA agents outside, defeated Widow, and tried to take her to the Avengers.
Strucker later sent Madame Hydra to Japan, where she made contact with a man named Gorgon and his Yashida Clan. Gorgon seemed willing to keep peace between his organization and HYDRA because he feared HYDRA's growing power.
Back on HYDRA Island, Madame Hydra gave her report to Strucker. At the same time, the Grim Reaper brought Hawkeye and Mockingbird as prisoners. Strucker wanted to drain Hawkeye of his life energy, but Madame Hydra stopped him, saying that having an Avenger as a captive could be useful.
Later, when Hawkeye and Mockingbird freed themselves and attacked them, Madame Hydra threw a grenade at Hawkeye. Luckily for those two, Captain America and Black Panther came to assist them in taking down HYDRA. Madame Hydra fought against Mockingbird, and almost shoot her, but she was interrupted by Hawkeye. After that, Mockingbird defeated Madame Hydra, and she turned into her Skrull form.
Madame Hydra's unconscious body was taken by the agents of S.H.I.E.L.D. to a secret location. During her captivity, her true Skrull nature was revealed. She is thought to be deceased since she hasn't awoken and remains in the same position as she was found.
